FLAG-NOA36 expression reproduces the endogenous intracellular localization of this evolutionary preserved protein. (A) Transfection of HeLa cells with the construct FLAG-NOA36. The expression detected with a specific anti-FLAG antibody by indirect immunofluorescence reproduces the nucleolar localization of the endogenous protein. Bar, 10 μm. (B) Western blot analysis of a transfection with the FLAG-NOA36 construct in HeLa cells. FLAG-NOA36 is detected by the anti-FLAG antibody in the transfected cell extract but not in the empty vector transfected one (left blot). A polyclonal anti-NOA36 antibody recognizes both, the endogenous NOA36 (white arrowhead) and the FLAG-NOA36 (black arrowhead) proteins (central blot and magnification bellow). The anti-FLAG magnetic beds successfully pull-down the FLAG-NOA36 recombinant protein (right blot)
